Removal of 3D animation

3D structures are derived, and should not appear without details of the source of the 3D structure coordinates, or, minimally, of the computations performed to generate the 3D structures from the 2D map of atoms and connectivities (line angle structure, as shown in infobox). Otherwise such 3D structures are unsourced WP:OR. This spinning 3D structure — with its inexplicably determined dihedral and bond angles — is such a case of unpublished, and undocumented original research. COX-2 selective or not?

Under mechanism of action:

... use of some COX-2 inhibitors with their adverse effects has led to massive numbers of patient family lawsuits alleging wrongful death by heart attack, yet other significantly COX-selective NSAIDs, such as diclofenac ...

It says that diclofenac in COX-selective. Does that mean COX-2 selective? That would seem to conflict this under cardiac side effects:

@146.255.183.82: Diclofenac isn't COX-2 selective, it inhibits both COX-1 and COX-2 like all the other conventional NSAIDs. This article looks significantly confused in this regard. From the introduction:
It blocks both COX-1 (like aspirin) and COX-2 (like celecoxib), and therefore is a COX-2 inhibitor.
COX-2 inhibitor usually means a COX-2 selective inhibitor, i.e. a 'coxib' like celecoxib. Non-selective COX inhibitors are usually referred to as (conventional) NSAIDs. In addition to this misnomer from the article, aspirin is not COX-1 selective, even though its COX-1 activity is more relevant at low doses. Aspirin significantly modifies COX-2, skewing its products towards lipoxins, rather than prostanoids.
I think I'm going to clear up the references to COX selectivity for now. The information should be accurate, and this point doesn't need to be overstated (especially in the introduction) given that diclofenac is so typical in this regard. AlexVojProc (talk) 13:19, 29 November 2022 (UTC)[reply]

diclofenac

Diclofenac was patented in 1965 by Ciba-Geigy

Don't see how this is possible as CIBA-Geigy did not exist in 1965. CIBA (acronym for Chemical Industries Basle should be in capital letters) CIBA and Geigy merged in 1970. I was there.

Molecular Interactions

It would be nice if there could be a subsection on the main article how diclofenac interacts with its target protein. The reason why this should be mentioned, IMO, is firstly because it is a small drug, second, it was historically important, but third and most importantly, there are .pdb structures showing it in a complex such as 1NR6.pdb, so it has a role that was already described (and can be studied in e. g. Too limited in the application described

Diclofenac is available in a lot more formulations than just Voltaren. There are eye drops, gels etc. I'm not expert enough to change the article. But someone should have a look at it and include the broader picture.
