Loss or Gain of function?

"Gain-of-function mutations in the intracellular NOD2 protein has been linked to increased risk for Crohn's disease." - Surprisingly, the predominance of evidence is that the Crohn's-associated mutations in NOD2 are LOSS-of-functions mutations. Specifically, cells transfected with wild-type NOD2 activate NFkB in response to the NOD2 agonist MDP; cells transfected with mutant NOD2 do not respond to MDP. A study just published (Kim et al. J Immunol 2011;187;2849-2852) seems to refute the experiment which suggested gain-of-function. Eaberry (talk) 03:53, 9 January 2012 (UTC)[reply]
