Summary

Description
English: Testosterone levels (ng/dL) during treatment with several progestins alone (weeks 0–2), then in combination with transdermal testosterone gel (50 mg/day) (weeks 2–6), followed by discontinuation and recovery (weeks 6–9) in 56 healthy young men (age 18–50 years). The progestins were as follows:
  • Cyproterone acetate (CPA) 10–20 mg/day orally (n=14)
  • Segesterone acetate (SGA; Nestorone) 2–3 mg/day transdermal gel (delivering 200–300 μg/day SGA) (n=14)
  • Norethisterone acetate (NETA) 5–10 mg/day orally (n=14)
  • Levonorgestrel (LNG) 120–240 μg/day orally (n=14)

In the progestin alone phase, changes in testosterone levels (from week 0 to week 2) were as follows:

  • CPA: ~431 ng/dL to ~149 ng/dL (–65%)
  • SGA: ~581 ng/dL to ~276 ng/dL (–52%)
  • NETA: ~527 ng/dL to ~231 ng/dL (–56%)
  • LNG: ~450 ng/dL to ~248 ng/dL (–45%)

The effects of the progestins on gonadotropin levels were not generally dose-related. This may be because the progesterone receptors for this effect were already saturated across the employed dose ranges. The lower and higher doses of the progestins were combined and averaged in the case of testosterone levels.
