English⧼Colon⧽ f6: Aberrant proplatelet formation by human megakaryocytes from patients with GPS.(a) Representative analysis of proplatelet formation and structure from control (CTRL)- (i–vii) and GPS-derived megakaryocytes by immunofluorescence (peripheral blood sample i–vi and viii–xiii; green = α-tubulin; red = CD61; blue = nuclei; scale bar = 50 μm) and light microscopy (bone marrow samples vii, xiv; scale bar = 20 μm). Pictures clearly show defective proplatelet elongation in GPS patients. (b) The percentage of proplatelet forming-megakaryocytes (ppf) was calculated as the number of megakaryocytes displaying at least one filamentous pseudopod with respect to total number of round megakaryocytes per analyzed field, and normalized with respect to CTRL. Data are presented as means ± SD (*p < 0.001). (c) Analysis of the number of proplatelet bifurcations per proplatelet forming-megakaryocytes (ppf). Data are presented as mean ± SD (*p < 0.001).
